For its third season, MLS Next Pro will feature 29 teams — 27 returning MLS affiliates and new independent teams Carolina Core FC and Chattanooga FC (formerly of NISA) — divided into four divisions:

Eastern Conference​
Northeast Division: Chicago Fire FC II, Columbus Crew 2. FC Cincinnati 2, New England Revolution II, New York City FC II, New York Red Bulls II, Philadelphia Union II, Toronto FC II. Southeast Division: Atlanta United 2, Carolina Core FC, Chattanooga FC, Crown Legacy FC, Huntsville City FC, Inter Miami CF II, Orlando City B.
Western Conference
Frontier Division: Austin FC II, Colorado Rapids 2, Houston Dynamo 2, Sporting Kansas City II, Minnesota United FC 2, North Texas SC, St. Louis City SC 2. Pacific Division: LA Galaxy II, Los Angeles FC 2, Portland Timbers 2, Real Monarchs, San Jose Earthquakes II, Tacoma Defiance, Whitecaps FC 2
